Protective Role of Sulfhydryl Compounds in Degradation of Collagen by Oxygen Free Radicals

Abstract
Sulfhydryl group containing compounds may protect oxidative injury to cellular components by drugs; mercury ions, ADP-chelated iron and X-ray irradiation through the sequestering effect on free radicals. In the present study, to explore the possibility for antiinflammatory effect of sulfhydryl compounds, effects of sulfhydryl compounds on degradation of collagen by interaction between Fe ` I and H2O2 were investigated. Their effects were studied with respect to collagen gelation, scavenging effect on reactive¢¥oxygen radicals and oxidation of sulfhydryl¢¥ groups of collagen. Inhibitory effect of Hz02 on collagen was stimulated by Fe¢¥ + in a dose dependent manner.. H2O2 stimulated autoxidation rate of Fe+ + Degradation of collagen caused by either interaction of Fe + + with H2O2 or xanthine/xanthine oxidase was effectively inhibited by scavengers of oxygen tree radicals and sulfhydryl compounds, mercaptopropionyl glycine, cysteine and glutathione. Both hydroxylation of sodium salicylate by interaction of Fe
with H2O2 (or xanthine and xanthine oxidase) and oxidation of DPBF by U.V. irradiation was inhibited by sulfhydryl compounds. Sul fhydryl compounds also inhibited oxidation of sulfhydryl groups of collagen by xanthine and xanthine oxidase. The results suggest that sulfhydryl compounds may have a scavenging effect on oxygen free radicals, particularly OH. or ¢¥02 and their effect could in part explain their antiinflammatory properties.
